摘 要:背景与目的:基因表达谱芯片是一种高通量,快速的检测基因表达量的手段。本研究利用这种新兴技术来研究鼻咽癌组织与鼻咽慢性炎症组织基因表达差异,以寻找鼻咽癌新的肿瘤相关候选基因和分子标志。方法:采用BioStarH-141s(2004)型含14112点的人类全长基因cDNA表达谱芯片对20例鼻咽癌组织及15例鼻咽慢性炎症组织进行检测,并用生物统计学及生物信息学方法分析它们之间的基因表达差异。结果:和鼻咽慢性炎症组织相比,20例放疗前鼻咽癌组织存在着大量的差异表达基因,涉及基因包括癌基因与原癌基因、免疫相关基因、细胞分裂、分化、增殖和凋亡相关基因、信号与蛋白传递相关基因、DNA结合转录和转录因子,一些生物酶的酶活调节等。结论:用基因表达谱芯片可以筛选出许多不同种类的参与鼻咽癌病变过程的重要基因,这为揭示鼻咽癌复杂的病变机理提供了研究的方向。Background and purpose: Gene expression profile is used to detect the gene expression level as a highthroughput and rapid method. In order to find the new candidate genes and molecular markers that are associated with nasopharyngeal carcinoma ( NPC), NPC relative gene has to be identified and distinguished from the genes in either other diseased or normal cells. This study is to focus on the screening genes by gene expression profile. Methods: Twenty NPC biopsies and fifteen nasopharynx chronic phlogistie biopsies were used to detect by BioStarH-141s(2004) profile gene chip which contains 14112 points of full length human genes. Biostatistics and bioinformaties were also used to analyse the differently expressed genes. Results: There were many different genes to be detected when gene profile for NPC were compared with the one for the normal tissue, with different genes including oneogenes and tumor suppressor genes, the genes that were associated with immunity, cell division, differentiation, proliferation, apoptosis, signal and protein transport, DNA binding and enzyme efficiency. Conclusions: Profile gene chip can be used to screen out many different kinds of genes that may participate in the development of NPC. The new technology may play an important role in the discovery of the mechanism of NPC pathological changes in the future.
