摘 要:目的观察预防性小脑顶核电刺激对自发性高血压大鼠(SHR)实验性脑梗死面积及脑组织内iNOS mRNA表达的影响。方法SHR持续性双侧顶核(FNs)电刺激60min,24h后行大脑中动脉栓塞术(MCAO),而对照组仅行MCAO,两组动物在脑梗死后不同时相处死,测定脑梗死面积,同时取脑组织标本送荧光定量PCR。结果MCAO后3h,FNs组梗死面积为(64.31±14.70)mm3,较MCAO对照组(84.77±21.01)mm3明显降低(P<0.05)。脑梗死后病灶半影区3、8、24h和3d时相点iNOS mRNA表达水平在FNs组分别为30.85±21.47,41.79±33.57,62.65±45.41和194.36±39.32均较MCAO组69.63±37.80,176.75±90.90,293.67±177.25和159.38±87.19明显降低(P<0.01)。从FNs后不同时间内对缺血性脑梗死的保护作用持续时间分析发现,在FNs后14d再行MCAO,其iNOS mRNA表达为248.43±60.70,与对照组无明显差异(P>0.05)。结论SHR预防性小脑电刺激可明显降低其后MCAO栓塞导致的脑梗死面积,该结果可能与有效抑制梗死半影区iNOS mRNA的表达有关。Objective To observe the effect of prophylactic electric stimulation of cerebella fastigia nucleus (FN) on experimental cerebral infarction size and iNOS mRNA expressions in cerebra in the rots. Methods The spontaneously hypertensive rats (SHRs) were given middle cerebral artery occlusion (MCAO) 24 h after a continuous 60 rain bilateral FNs electric stimulation, while the control group received only MCAO. All animals were killed at different time points after cerebra] infarction to detect the volume of infarction size and the expression of iNOS mRNA in penumbra of cerebral ischemia by the techniques of fluorescence quantitative RT-PCR. Results The volume of infarction size in FN group [ (64. 31 ± 14. 7 ) mm^3 ] was lower than that in MCAO control group [ (84. 77 ± 21.01 ) mm^3 ] (P 〈 0. 05 ) 3 h after MCAO. The expression levels of iNOS mRNA in penumbra] region in cerebral infarction lesion at 3, 8, 24 h and 3 d in FN group (69.63 ± 37.8, 176.75 ± 90.9,293.67 ± 177.25, 159.38 ± 87. 19 ) were significantly lower than those in MCAO group (69.63 ± 37.8, 176.75 ± 90. 9,293.67 ± 177. 25, 159.38 ± 87. 19 ) ( P 〈 0. 01 ). The analysis on the duration of protective effect of FN electric stimulation in different time on ischemia cerebra] infarction showed that MCAO 14 d after FN electric stimulation with iNOS mRNA expressing 248. 43 ± 60. 70 had no obvious difference from MCAO control ( P 〉 0.05 ). Conclusiona FN electric stimulation could effectively reduce ischemic lesion caused by MCAO in SHRs, which might be related to the inhibition of iNOS mRNA expressions in penumbral region in cerebra] infarction lesion.
