摘 要:目的研究耐药性颞叶癫痫患者脑组织中细胞分裂周期蛋白42(Cdc42)的表达,探讨其在耐药性颞叶癫痫形成中的意义。方法在重庆市癫痫病防治中心建立的耐药性颞叶癫痫患者术后脑组织库中抽取20例患者的脑组织,用免疫组织化学、免疫荧光分别检测其中 Cdc42的表达,并与性别相同、年龄近似的对照组进行比较。结果免疫组织化学检测到病例组、对照组颞叶,病例组海马、对照组海马平均吸光度(A)值分别为0.038±0.016,0.020±0.005,0.049±0.009,0.025±0.009,t 检验提示病例组与对照间差异有统计学意义(P<0.01)。免疫荧光法观察病例组、对照组颞叶,病例组、对照组海马荧光强度分别为18.3±2.5,11.8±2.9,23.8±3.3,10.1±2.6,病例与对照组间同样存在统计学差异(P<0.01)。结论 Cdc42在耐药性颞叶癫痫患者的颞叶皮质及海马中表达增强,提示其参与了耐药性颞叶癫痫的形成。Objective To investigate the expression of cell division cycle 42 GTP-binding protein (Cdc42) in the brain tissue of patients with intractable temporal epilepsy and discuss its effect on epileptogenesis. Methods Twenty samples brain tissues were collected from the brain tissue bank of intractable temporal epilepsy established by Epilepsy Preventive and Therapeutic Center of Chongqing. The expression of Cdc42 was determined by immunohistochemistry and fluomimmunoassay. Twenty brain tissue samples were collected from autopsy on 20 sex and age-matched persons who died from accident were used as controls. Results The absorption values of Cdc42 in the temporal lobe of the patients was 0.038 ± 0.016, significantly higher than that of the controls (0.020 ±0. 005 ,t = 14.59 ,P 〈0.01 ). The absorption values of Cdc42 in the hippocampus of the patients was 0. 049 ±0. 009,t = 12.01 ,P 〈0.01 ). Fluomimmunoassay showed that the fluorescence intensity of Cdc42 in the temporal lobe of the patients was 18. 3 ± 2. 5, significantly higher than that of the controls ( 11.8 ± 2.9, t = 33.77, P 〈 0.01 ) , and the Fluorescence intensity of Cdc42 in the hippocampus of the patients was and control hippocampus were 23. 8 ± 3.3, significantly higher than that of the controls ( 10. 1 ± 2.6, t = 12. 01, P 〈 0. 01 ). Conclusion The overexpression of Cdc42 in the cortex of temporal lobe and hippocampus may play an important role in the epileptogenic mechanisms of intractable epilepsy.
