摘  要:目的探讨5-氨基乙酰丙酸-光动力疗法(5-aminolevulinic acid-mediated photodynamic therapy,ALA-PDT)在急性淋巴细胞白血病(ALL)小鼠骨髓移植后对移植物抗宿主病(GVHD)和移植物抗白血病作用(GVL)的影响。方法以ALL小鼠为模型,经致死剂量60Co照射后,对其进行同种异基因骨髓移植的同时,静脉输注经ALA-PDT处理的供受鼠混合淋巴细胞,同时设立骨髓细胞加脾细胞移植对照组及空白对照组。观察骨髓移植后受鼠的一般情况、30 d生存率、造血功能恢复情况以及肝脏等组织的病理学改变。结果ALA-PDT处理组小鼠30 d生存率明显高于其他各组,差异有统计学意义(P<0.01);其造血功能恢复情况及GVHD反应均较其他各组有所改善。结论ALA-PDT能明显减轻小鼠骨髓移植后GVHD反应,并保留一定的GVL作用。Objectives To elucidate the effects of 5-aminolevulinic acid-mediated photodynamic therapy (ALAPDT) on graft-versus-host disease (GVHD) and graft-versus-leukemia (GVL) effect following allogenic bone marrow transplantation (allo-BMT) in mice with acute lymphoblast leukemia (ALL). Methods Allo-BMT was carried out in ALL mice following lethal dose of 60^Co radiation; meanwhile, mixed lymphocytes prepared by ALA-PDT were infused into ALL mice. Bone marrow cells-spleen cells transplantation group and radiated mice without any intervention were used as controls. General condition, survival rate on day 30, recovery of haematopoietic function, and pathological changes of the liver were applied to evaluate the beneficial effects of ALA-PDT on GVHD and GVL in ALL mice. Results ALA-PDT pretreatment significantly (P 〈 0.01) improved the survival rate of ALL mice. Recovery of haematopoietic function and GVHD development are more improved than that of the other two control groups. Conclusions ALA-PDT significantlyalleviated GVHD in ALL mice following allo-BMT and reserved its GVL effect.
