摘 要:目的探讨微泡造影剂声诺维(SonoVue)联合超声辐照对RPE细胞增殖的影响。方法在96孔板中,对照组不处理,实验组分为超声声强、辐照时间、造影剂浓度、超声形式组处理,MTT法检测对RPE细胞增殖的影响,计算细胞存活率。结果实验组的吸光度值较对照组有不同程度的降低,各组间差异有显著性意义(P=0.0001)。第4、6、8、9、10组与第1组比差异有显著性意义(P<0.05)。造影剂浓度增加对细胞增殖有明显抑制作用(P<0.05)。结论一定剂量的微泡造影剂联合超声辐照对RPE细胞增殖有抑制作用。Objective To investigate whether ultrasound mediated microbubbles destruction could impact on the proliferation of cultured retinal pigment epithelium (RPE) in vitro. Methods RPEs were cultured in 96 wells and exposed to ultrasound in different intensity, forms, radiation and different concentration of SonoVue in the experimental groups, but no treatment was applied to the control group. RPE proliferation was observed on MTT assay and the cell survival rate were calculated. Results The absorbed optical density of the experimental group was lower than that of the control group and the difference in the intergroups was significant (P = 0. 0001 ). The difference of 4, 6, 8, 9, 10 vs the first were significant (P〈0. 05) . The increase of contrast agent concentration would inhibit cell proliferation obviously (P〈0. 05) . Conclusions Ultrasound mediated microbubble destruction could impact on the proliferation of RPE.
