摘 要:目的:观察复方奥硝唑甲磺酸培氟沙星缓释牙栓对体外培养的人牙周膜成纤维细胞(HPDLFS)生长的影响。方法:采用组织块法培养HPDLFS,观察缓释牙栓在0、0.1、0.5、2.5、10、50、200g/L对体外培养HPDLFS蛋白合成及DNA合成的影响,MTT法检测细胞在6、12、24、48、72h光密度值(OD),计算细胞相对增殖率(RGR)。结果:免疫组化试验证实;不同浓度缓释牙栓对HPDLFS蛋白合成及DNA合成与对照组相比无显著性差异(p>0.05);随作用时间、浓度不同,HPDLFS生长趋势稳定,RGR均在90%以上。结论:在200g/L缓释浓度范围内,该缓释牙栓有较佳的生物相容性,未显示有细胞抑制作用。Objective:To evaluate the effects of compound omidazole and pefloxacinmesylat sustained-release periodontal suppository on to human periodontal ligament fibroblasts (HPDLFS). Method:The method was employed to culture HPDLFS.The total amount of protein was detected with Counmassie Bringt Blue method and DNA synthesis was measured by 3H-TdR, It was observed in doses (0.0.1.0.5.2.5.10.50.200 g/L) to HPDLFS: MTT colorimetry was used to evaluate on different time (6.12.24.48.72 h) to cellular OD and relevant analysis was done through reckoning cellular RGR. Result:HPDLFS were identified by cytochemistry,It was not effected that the Counmassie Bringt Blue method and DNA synthesis in different doses (P〉0.05), HPDLFS is growth with lasting tendency, RGR〉90% .Conclusion: The results indicated that within proper doses Periodontal Suppository (200 g/L)had excellent biocompatibility, It had not restraining effect to HPDLFS.
