摘  要:从污泥堆肥中分离得到一株具有溶胞能力的嗜热菌SY-9,经形态及16SrDNA测序初步鉴定为土芽孢杆菌属(Gebacillus sp.).对其生长、产酶及溶胞特性进行了研究.结果表明,SY-9最适生长pH值为7-9,最适生长温度约60℃,60℃的世代时间为34min.SY-9培养上清液具有溶胞能力,上清液经过热处理后溶胞能力明显下降,说明溶胞能力主要来自酶的作用.SY-9间歇培养过程中,培养上清液对大肠杆菌(Escherichia coli)的溶胞活性在SY-9进入稳定生长期后逐渐升高,达到最大值后随着培养时间的延长逐渐下降.SY-9培养上清液对受试的5株革兰氏阴性菌(3株大肠杆菌及2株假单胞菌)及部分革兰氏阳性菌(枯草芽孢杆菌、红球菌、球形节杆菌及溶壁微球菌)都具有溶胞能力.One thermophilic bacterial strain SY-9, isolated from compost of municipal sludge, showed significant capability to lyse bacterial cells. The strain was identified as Geobacillus sp. basing on morphological characteristics and homology identification of 16S rDNA sequence. The optimal growth pH of SY-9 was 7-9. The optimal growth temperature was about 60℃ with a generation time of 34min. The culture supematant of SY-9 possessed lytic activity, after thermal treatment, the lytic activity decreased significantly, indicating the lytic activity came from the action of enzymes. In the batch culture process of SY-9, the lytic activity of culture supematants on Escherichia coli increased gradually after SY-9 entered the stationary growth phase, and then decreased gradually with prolonging of culture time after reaching the greatest value. The culture supematanf of SY-9 showed lytic activity on the tested five Gram-negative strains (three Escherichia coli strains and two Pseudomonas sp. strains) and partial Gram-posftive strains (Bacillus subtilis, Rhodococcus sp., Arthrobacter globiformis and Micrococcus lysodeikticus).
