恶性纤维组织细胞瘤Cath-D mRNA表达的研究

摘　　要：目的探讨组织蛋白酶D(Cathepsin D,Cath-D)mRNA在恶性纤维组织细胞瘤(malig-nant fibrous histiocytoma,MFH)组织中的表达及意义。方法应用原位杂交的方法检测65例MFH组织中Cath-D mRNA的表达情况,同时选取10例正常纤维组织做对照。结果在10例正常纤维组织中Cath-D mRNA均不表达;Cath-D mRNA在肿瘤组织中的阳性表达率69.2%;Ⅰ-Ⅱ期阳性率为50.0%,Ⅲ-Ⅳ期阳性率为85.7%,两者相比有显著性差异(P<0.01);Cath-D mRNA的阳性表达率与病理分化程度和生存期显著相关(P<0.05)。结论Cath-DmRNA的过表达与恶性纤维组织细胞瘤浸润、转移相关,而且可能是预测恶性纤维组织细胞瘤预后的生物学指标。Objective To study the mRNA expression and significance of Cathepsin D （Cath-D） in malignant fibrous histiocytoma （MFH）. Methods 65 MFH tissues and 10 normal fibrous tissues were exam- ined for Cath-D mRNA by the method of in-situ hybridization. Results Cath-D mRNA did not express in 10 normal fibrous tissues. The mRNA expression of Cath-D in MFH tissues were 69.2%. In stage Ⅰ - Ⅱ , Cath-D mRNA positive rate was 50.0% ,which was markedly lower than those in stage Ⅲ -Ⅳ（85.7%）.There were statistically significance（ P 〈 0.01）. The positive expression rate of Cath-D mRNA was significant associated with degree of patholdgical differentiation and life span （ P 〈 0.05）. Conclusion The over expression of Cath- D mRNA have a close relationship with the invasion and metastasis of MFH, which suggest that it is an useful prognostic indicator for MFH.
