摘  要:目的:研究王不留行Vaccaria segetalis(Neck)Garcke(VSG)水煎剂对主动脉环的舒张作用及机制。方法:采用家兔离体主动脉平滑肌标本,以去甲肾上腺素(NA)预收缩后,给予不同剂量的VSG观察其张力变化,并探讨去除血管内皮、给予NO合酶抑制剂左旋硝基精氨酸(L-NNA)、甲烯蓝(MB)、环氧酶抑制剂吲哚美辛和β肾上腺素能受体阻断剂普萘洛尔对血管张力变化的影响。结果:VSG对血管环静息张力无明显影响,但不同剂量的VSG可使10-6mol/L NA预收缩的血管产生明显舒张(r=0.76,P<0.001)。去除血管内皮、10-4mol/L L-NNA、10-5mol/LMB或吲哚美辛可减弱VSG的舒张作用,但普萘洛尔无影响。另外,2mg/ml VSG温育无内皮的血管环后,对NA和KCl的量效曲线无明显影响。结论:VSG舒张主动脉的作用是内皮依赖性的,且与其释放的NO有关;也可能是通过PGI2/cAMP介导,降低胞浆内钙离子引起血管平滑肌舒张。Objective: To study the vasodilation effects of the water decoction of Vaccaria Segetalis (Neck) Garcke (VSG) on rabbit aortic rings and to discuss the possible involved mechanism. Methods: We investigated the response to different concentration of VSG in isolated rabbit's aortic rings precontracted with noradrenaline (NA). Then we assessed the effects on denuded endothelium, coincubated with nitric oxide synthase inhibitor Nw-nitro-L-arginine (L-NNA), methylene blue (MB), prostaglandin synthase inhibitor and blockage of the adrenergic β-receptor, propranolol on VSG-induced relaxation. Results: VSG had no effects on the resting tension of rabbit aortic rings. However, it induced a significantly concentration-dependent relaxation on NA (10-6mol/L) precontracted rings (r = 0.76, P 〈 0.001), which could be reduced by denuding endothelium or by 10^-4mol/L L-NNA, 10^-5mol/L MB and prostaglandin synthase inhibitor. But propranolol, the blockage of the adrenergic β-receptor, showed no effects. Furthermore, the concentration-response curves of NA and KCI had not been shifted with VSG (2mg/ml) on the rabbit aortic rings denuded endothelium. Conclusion: There can be seen an endothelium-dependent vasorelaxation on the isolated rabbit's aortic rings with VSG, and nitric oxide (NO) might be involved in. The signal transduction pathway PG12-cAMP-iCa + also might be involved in.
