胃癌患者外周血中骨桥蛋白和基质金属蛋白酶-9的含量分析及临床意义

摘　　要：目的 探讨骨桥蛋白（OPN）和基质金属蛋白酶-9（MMP-9）在胃癌患者外周血中的含量变化及其临床意义.方法 采用双抗体夹心ABC-ELISA法检测51例胃癌患者、29例慢性萎缩性胃炎患者、31例健康者和29例胃癌术后6～12个月随访者外周血中的OPN和MMP-9的含量;应用免疫组织化学方法检测28例胃癌组织中OPN和MMP-9的表达.结果 胃癌患者外周血中OPN和MMP-9的含量明显高于健康者和慢性萎缩性胃炎;胃癌患者外周血中OPN和MMP-9的含量与胃癌的临床分期及预后明显相关,而与性别、年龄、肿瘤大小及分化程度无关;胃癌组织中OPN和MMP-9高表达,外周血中OPN和MMP-9存在相关性.结论 检测外周血中OPN和MMP-9的含量对胃癌的诊断、治疗、预后评估及病情监测有重要意义.Objeclive To investigate the plasma level and clinical significance of osteopontinand （OPN） and matrix metalloproteinase-9 （MMP-9）in patients with gastric cancer. Methods Plasma level of osteopontin and matrix metalloproteinase-9 were detected by ABC-ELISA method in 51 patients with gastric eaneer、29 patients with chronic atrophic gsstritis、 31 normal peoples and 29 patients with gastric cancer cut away;The expression of OPN and MMP-9 was examined by immunohistoehemistry method. Results The plasma leverlevel of OPN and MMP-9 in gastric cancer group was higher than that in normal group or chronic atrophic gastritis. The plasma leverlevel of OPN and MMP-9 in gastric cancer patients was correlated with clinical stage and prognosis, but not correlation with sex, age, size or differentiated degree of tumour;Preoperative and postoperative plasma levels of OPN and MMP-9 in gastric cancer group were obviously different;There were also some correlations between OPN and MMP-9 plasma level. Conclusion To detect Detection of the plasma levels of OPN and MMP-9 in gastric cancer patients has greatly clinical significance in the treatment diagnosis prognois evaluation and monitoring for gastric cancer patients.
