摘  要:【目的】探讨注意缺陷多动障碍(attention-deficit hyperactivity disorder,ADHD)动物模型SHR(spontaneously hypertensive rats)大鼠与对照组WKY(Wistar-Kyoto rats)大鼠前额叶组织基因表达谱差异,试图从基因网络角度全面理解ADHD的分子机制及差异表达基因与其生物学特征间的关系。【方法】提取SHR/WKY大鼠前额叶组织mRNA,反转录成cDNA,体外转录并生物素标记cRNA,片断化后,分别与Affymetrix公司制备寡核苷酸RAE230A基因芯片杂交,对二者差异表达基因进行生物信息学分析。【结果】SHR大鼠与对照组WKY大鼠前额叶皮层组织中差异表达2倍以上的探针组数209个。对代表已知功能的67个独立基因进行分类,结果发现差异表达基因以神经发育相关、代谢相关、信号转导、免疫相关、转录调节相关基因为主,同时还涉及突触可塑性、髓鞘形成、学习记忆、高血压、细胞凋亡等不同层次、不同功能的基因群。【结论】SHR大鼠前额叶皮层组织的基因表达谱存在神经发育相关基因的表达异常、免疫相关基因的表达下调、多个转录因子的普遍上调和部分单胺类神经递质相关基因表达上调等特征。[Objective] To investigate the difference in gene expression profiles between the prefrontal cortex of spontaneously hypertensive rats (SHR) and Wistar-Kyoto rats (WKY) by using microarray analysis, and to analyze the feature of gene expression profiles in the prefrontal cortex(PFC) of SHR . [ Methods] The total RNA was extracted from the prefrontal cortex of SHR rats and WKY and gene expression was analyzed using Affymetrix oligonucleotide microarray technology (RAE230A). The functions of the differentially expressed genes were analyzed by bioinformatics. [ Results] 209, 60, 23 probe sets genes were found significantly differentially expressed over 2, 4, 8 folds respectively between the prefrontal cortex of SHR and WKY. Among the 67 unigenes with known function in the differentially expressed genes,different clusters of genes associated with neuro development, metabolism, signal transduction, DNA transcription, immune, etc were concerned. [Conclusion] Dysregulation of synaptic transmission, impaired the formation of the mylin of axons, upregulation of DNA transcription related genes, decreased immunity related genes etc are the features of gene expression profiles of the prefrontal cortex of SHR.
