摘  要:目的:研究种间胚胎植入期母体淋巴结中表达CD4+、CD8+、CD25+、CD69+T细胞的百分比变化,并探讨这种变化对种间胚胎植入的影响。方法:利用荧光标记的单克隆抗体染色结合流式细胞术,检测种间、同种胚胎移植以及同时期假孕母体淋巴结中表达CD4+、CD8+、CD25+、CD69+的T淋巴细胞对应CD3+细胞的百分率。结果:与同种胚胎移植后的植入相比,种间胚胎移植后植入期母体淋巴结中,CD3+CD4+、CD3+CD25+淋巴细胞占CD3+淋巴细胞的比例均极显著低于同种胚胎移植的植入期受体(P<0.01);而CD3+CD8+淋巴细胞占CD3+淋巴细胞的比例显著低于同种胚胎移植的植入期受体(P<0.05);CD3+CD69+淋巴细胞在种间、同种胚胎植入期母体淋巴结中的比例却无显著差异(P>0.05)。种间胚胎植入时母体淋巴结中CD4+/CD8+的比例,与同种胚胎植入受体和假孕小鼠相比,三者之间均无统计学意义上的显著差异(P>0.05)。结论:种间胚胎移植后从植入期开始,母体淋巴结中的淋巴细胞就发生了不利于胚胎植入的全身免疫反应。To study percentage differences of CD4^+、CD8^+、CD25^+、CD69+T lymphocytes in maternal lymph nodes derived from the recipients of interspecific, intraspecifc embryo transfer during implantation and from pseudopregnant mice at corresponding time, and to explore the effects of these differences on interspecific embryo implantation. Methods: Fluorescence conjugated monoclonal antibodies and flow cytometry were employed to detect the percentages of CD4^+、CD8^+、CD25^+、CD69+ compared to CD3^+ expressive lymphocytes in maternal lymph nodes derived from recipients of interspecific, intraspecifc embryo transfer during implantation and from pseudopregnant mice at corresponding time. Results:Compared to embryo implantation after intraspecific embryo transfer, the percentages of CD3^+CD4^+ and CD3^+CD25^+ cells in CD3^+ lymphocytes of lymph nodes derived from recipients of interspecific embryo transfer were both very significantly lower, respectively ( P 〈 0. 01 ). While the percentage of CD3^+CD8^+ cells in CD3^+ lymphocytes was significantly lower than that of intraspecific embryo transfer( P 〈 0. 05 ) , the percentage of CD3^+CD69+ cells in CD3^+ lymphocytes had no significant difference to that of intraspecific embryo transfer(P 〉0. 05). During embryo implantation, it had no significant differences among the ratios of CD3^+CD8+lymphocytes in maternal lymph nodes in all three experimental groups( P 〉 0. 05). Conclusion : Systemic immunological changes harmful to embryo implantation by lymphocytes in lymph nodes derived from recipients of interspecific embryo transfer were happened early even from the time of embryo implantation.
