摘  要:目的:探讨外源性降钙素基因相关肽(Calcitonin gene related peptide,CGRP)对局灶性脑缺血再灌注大鼠顶叶皮质tau蛋白过度磷酸化的影响。方法:用线栓法制作局灶性脑缺血再灌注模型,应用免疫组织化学SABC法、Western Blotting和图像分析方法检测大鼠顶叶皮质tau蛋白在Ser199/202位点磷酸化程度和总tau蛋白表达、以及CGRP对缺血神经元tau蛋白过度磷酸化的影响。结果:缺血再灌注顶叶皮质tau蛋白在Ser199/202位点磷酸化水平和总tau蛋白显著升高(P<0.05);CGRP组大鼠顶叶皮质tau蛋白在Ser199/202位点磷酸化水平显著低于缺血再灌注组,总tau也降低(P<0.05)。结论:CGRP明显减轻局灶性脑缺血再灌注大鼠顶叶皮质tau蛋白磷酸化程度,降低tau蛋白磷酸化水平可能对缺血神经元起保护作用。Objective:To explore the effect of exogenous calcitonin gene-related peptide (CGRP) on tau hyperphosphorylation in rat parietal cortex during focal cerebral ischemia/reperfusion (I/R). Methods:The rat model of focal cerebral I/R was established by occluding the right middle cerebral artery with intraluminal suture method. The levels of tau hyperphosphorylation at Ser199/202 site and total tau protein in the rat parietal cortex were detected by immunohistochemical SABC method and Western blotting. The immunoreactive positive products were analyzed by image analysis system. Results:The levels of tau hyperphosphorylation at Ser199/202 site and total tau protein in the rat parietal cortex were significantly higher in I/R group than in sham operation group (P 〈 0.05). CGRP significantly inhibited tau hyperphosphorylation at Ser199/202 site,and the level of total tau protein was significantly lower in CGRP group than in I/R group (P 〈 0.05). Conclusion:CGRP can decrease the level of tau hyperphosphorylation in the parietal cortex of focal cerebral I/R rat,and the inhibition of tau hyperphosphorylation may be involved in protecting the ischemic neurons.
