川芎中阿魏酸、川芎嗪对小鼠神经系统的影响

摘　　要：目的:研究川芎中阿魏酸、川芎嗪对中枢神经系统的影响。方法:采用镇静催眠实验,腹腔分别注射320,160,80mg.kg-1阿魏酸及2000,1000,500mg.kg-1川芎嗪,观察用药前后对小鼠旷场行为和自主活动的影响,及其协同戊巴比妥钠对小鼠的催眠作用影响。结果:各剂量组的川芎嗪和阿魏酸在给药60,90,120min后都能引起小鼠自主活动的减少(P<0.01)。2000,1000mg.kg-1川芎嗪使戊巴比妥钠睡眠时间延长(P<0.01,P<0.05),能加强阈下剂量戊巴比妥钠的催眠作用。500mg.kg-1川芎嗪和阿魏酸各剂量组对戊巴比妥钠阈下催眠均无明显影响(P>0.05)。结论:川芎组分川芎嗪及阿魏酸对中枢神经系统具有一定的镇静、抑制作用,且较高浓度的川芎嗪与戊巴比妥钠具有明显的协同催眠效果。OBJECTIVE To study effects of different components of Ligusticum chuanxiong Hort of ferulic acid and ligustrazine on the nervous system. METHODS Using the sedative and hypnotic experiments to observe the open-field activity and the spontaneous activity after mice were intraperitoneally injected 320,160,80 mg· kg^-1 of ferulic acid and 2 000, 1 000, 500 mg · kg^- 1 of ligustrazine , measuring the changes of hypnotic action induced with pentobarbital in subthreshold. RESULTS After 60,90,120 min intraperitoneally injection of ligustrazine （every concentration） and ferulic acid （every concentration）, the rates of spontaneous activity were inhlbited（P〈0. 01 ）, After intraperitoneally injection 2 000, 1 000 mg· kg^-1 of ligustrazine the sustained period of sleep lengthened（P〈0. 01 .P〈0. 05）, strengthen action of hypnotic action induced with pentobarbital in subthreshold, but after intraperitoneally injection of 500 mg·kg^-1 of ligustrazine and ferulic acid（every concentration） ,the rates hypnotic of the pentobarbhal in subthreshold have no changes （P〈 0. 05 ）. CONCLUSION The components of Liguticum chuan.ziong Hort of ligustrazine and fenulic acid was shown to affect the nervous center system evidently in sedative and inhibition. High concentration of ligustrazine has obvious hypnotic effect in conjunction with sodium pentobarbhal.
