摘 要:目的探讨组合染色方法用于过敏性猝死豚鼠肥大细胞脱颗粒现象的可行性及其在法医学鉴定中的作用和意义。方法18只豚鼠,雌雄不限,体重200g左右,随机分为实验组和对照组。实验组采用多人混合血清注射液致敏豚鼠,建立过敏性猝死的动物模型,取豚鼠肺组织,应用BB-IG-MY组合染色法显示肥大细胞脱颗粒现象。对照组豚鼠用生理盐水代替多人混合血清,其余处理同实验组。结果实验组豚鼠在被抗原再次攻击后20s左右出现活动减少,1min左右出现休克症状,2-5min死亡,死亡率100%。对照组豚鼠均未出现任何症状。组合染色法结果表明,肥大细胞胞质颗粒呈棕色,细胞核呈绿色,红细胞呈黄色。过敏性猝死豚鼠肥大细胞脱颗粒现象明显,而对照组豚鼠肥大细胞未见脱颗粒现象。结论过敏性猝死时,肥大细胞有明显的脱颗粒现象。在实际法医学鉴定中,肥大细胞脱颗粒可有力提示过敏性疾病的存在。Objective To explore the feasibility of combined-staining for observing mast cell degranulation in guinea pigs with sudden death due to anaphylactic shock,and to investigate its role in the medicolegal expertise.Methods Eighteen guinea pigs were randomly divided into experimental group and control group.The guinea pigs were injected with pooled serum to establish anaphylactic shock model in experimental group.Mast cell degranulation in lung was observed by BB-IG-MY combined-staining.In control group,the guinea pigs were injected with physiological salt solution.Results In experimental group,the activities decreased at 20 s after second sensitization,anaphylactic shock appeared at 1 min,and all the guinea pig died in 2-5 min.Mast cells had released the granules in guinea pigs with sudden death due to anaphylactic shock,but not in control group.Conclusion Mast cells have released the granules in guinea pigs with sudden death due to anaphylactic shock.In the medicolegal expertise,mast cell degranulation could imply anaphylactic shock.
