摘  要:目的研究环氧合酶-2mRNA(COX-2mRNA)在骨肉瘤中的表达及其与CD105mRNA的相关性。方法应用原位杂交的方法检测人骨肉瘤中COX-2mRNA和CD105mRNA的表达,并计算CD105mRNA标记的微血管密度(MVD)。结果COX-2mRNA在骨肉瘤中表达阳性率为77.5%,COX-2mRNA的表达与外科分期和肿瘤的转移密切相关(P<0.05);外科分期的I期与Ⅱb期、Ⅲ期间的MVD值的差异有统计学意义(P<0.05),而I期与Ⅱa期、Ⅱa期与Ⅱb期之间的差异无统计学意义(P>0.05);转移组的骨肉瘤其MVD值高于未转移组,二者之间的差异有统计学意义(P<0.05);COX-2mRNA表达与MVD值呈正相关(P<0.05)。结论COX-2mRNA在骨肉瘤组织中呈现高表达,其高表达可能促进了骨肉瘤新生血管的生成。Objective To study the expression of cyclooxypenase-2mRNA (COX-2mRNA) and the correlation between COX-2mRNA and CD105mRNA in osteosarcoma. Methods The expression of COX- 2mRNA and CD105 mRNA in human osteosarcoma was detected with in-situ hybridization. Then, in order to count the microvessel density (MVD), cells were marked with CD105mRNA. Results The positive rate of COX-2mRNA expression was 77.5%. The expression of COX-2 mRNA was positively correlated with surgical grade and metastasis of osteosarcoma. There were significant differences between Stage I and IIb, Stage I and III (P 〈 0.05). No significant difference was found between Stage I and IIa, Stage IIa and IIb (P 〉 0.05). Metastasis cases had higher MVD than in the cases without metastasis (P 〈0.05). The positive correlation was found between expression level of COX-2mRNA and MVD marked by CD105mRNA. Conclusion The overexpression of COX-2mRNA is found in human osteosarcoma. Therefore, angiogenesis in osteosarcoma may be promoted by COX-2.
