摘 要:目的比较人喉鳞癌放射抗拒细胞株Hep-2R与其亲本细胞株Hep-2的体外增殖、运动及侵袭能力。方法分别培养人喉鳞癌放射抗拒细胞株Hep-2R和Hep-2,通过克隆形成实验测定其增殖能力,Transwells小室法测定运动及侵袭能力。结果Hep-2和Hep-2R的克隆形成率分别为(20.75±1.29)%和(19.17±0.82)%,二者有显著性差异(P<0.05);运动力(6个镜下视野的穿膜细胞数)分别为(122.0±6.8)和(117.0±5.8),二者无显著性差异(P>0.05);侵袭力(6个镜下视野的穿膜细胞数)分别为(166.0±9.8)和(166.4±8.4),二者无显著性差异(P>0.05)。结论人喉鳞癌放射抗拒细胞株Hep-2R较其亲本细胞株Hep-2的体外增殖能力弱,而运动和侵袭能力无显著性差异。Objective To compare the abilities of proliferation, mobility and invasion between radioresistant human larynx squamous carcinoma cell line Hep-2R and its parent cell line Hep-2 in vitro. Methods The above-mentioned cell lines were cultured. The ability of proliferation was measured with colony formation. Cell mobility and invasion were assayed by transwell chamber model assay. Results The colony formation rates of the two cell lines were(20.75±1.29)% and(19.17±0.82)%, P〈0.05. Transwell chamber model assay showed the cell mobility abilities were (122.0±6.8) and(117.0±5.8), P〉0.05; cell invasion abilities were(166.0±9.8) and(166.4±8.4), P 〉0.05. Conclusion The ability of proliferation in radioresistant human larynx squamous carcinoma cell line Hep-2R was weaker than in its parent cell line Hep-2, but there were no significant differences in mobility and invasion abilities between these two cell lines.
