兔血管内皮细胞对角膜后弹力层代谢的影响

摘　　要：目的:检测移植后生长于角膜内皮面的自体血管内皮细胞对角膜后弹力层代谢的影响。方法:家兔26只随机分成3组:实验组10只,将培养的自体血管内皮细胞移植到无后弹力层的角膜内皮面;实验对照组10只,移植无角膜内皮层的自体植片;空白对照组6只,移植带角膜内皮细胞的自体植片,术后观察3组角膜植片的水肿变化。术后15d,取下各组角膜植片做病理切片HE染色观察各组植片的角膜后弹力层变化及其上的细胞分布情况。结果:实验组角膜植片的内皮面长满培养的自体血管内皮细胞。病理切片显示,生长的细胞呈单层分布,内皮面未见明显的后弹力膜样物质形成;实验对照组角膜植片高度增厚,内皮面无细胞生长和后弹力膜样物质形成;空白对照组角膜植片内皮面可见明显的后弹力层,其上可见单层细胞分布。结论:血管内皮细胞具有一定的角膜内皮细胞样功能,但不能产生角膜后弹力层样物质。AIM： Detecting influence of autogeneic vascular endotheliocytes growing over posterior corneal surface after transplantation on metabolism of Descemet membrane. METHODS： 26 experimental rabbits were divided into the following three groups： the experimental group （10 rabbits）, the experimental control group （10 rabbits） and the blank control group （6 rabbits）. In experimental group, cultivated autogeneic vascular endotheliocytes would be grafted on corneal endothelium without Descemet membrane, in the blank control group, autogenous graft with corneal endothelium cells would be transplanted directly to the contralateral eyeball for contrasting observation with other two groups; in the experimental control group, all the autogeneic grafts whose Descemet membranes were removed from corneal stroma layer would be sutured to the corneal bed of the contralateral eyeball for the same purpose as in blank control group. Corneal transparence of all animals was frequently observed during the postoperative 15 days. An ultrasound apparatus was adopted to measure the postoperative thickness variability of corneal grafts every 7 days in each group. On the postoperative 15th day, corneal grafts of all groups were taken out of their left eyeball and put under the inverted phase contrast microscope to observe the distribution state of vascular endotheliocytes on posterior corneal surface without Descemet membrane, and pathologic sections and H-E staining were carried out for detection of Descemet membrane metabolism in each group. RESULTS： The clarity of corneal graft in the experimental group had an advantage over that in the experimental control group, but inferior to that in the blank control group. Thickness values of corneal grafts in each group were analyzed with T test and the results are as follows： thickness difference of the grafts was scarcely manifest between the experimental group and blank control group, but corneal thickness of the latter was especially similar to the normal corneal thickn
