摘  要:目的研究骨形态发生蛋白(BMP)-2和BMP-6对人骨肉瘤细胞株MG63和U2OS的作用。方法分别用重组腺病毒AdBMP-2、AdBMP-6和AdGFP感染骨肉瘤细胞MG63和U2OS,利用台盼蓝拒染法、TUNEL法、吖啶橙/溴乙啶(AO/EB)荧光双染法、Transwell小室和碱性磷酸酶(ALP)活性测定法分别检测细胞增殖、凋亡、迁移以及成骨分化能力的变化。结果与感染AdGFP的相应骨肉瘤细胞株(对照组)相比,分别感染AdBMP-2和AdBMP-6的两种骨肉瘤细胞株的细胞存活率随时间延长逐渐降低,凋亡率逐渐增加,细胞穿膜数逐渐减少,ALP活性逐渐增加。结论BMP-2和BMP-6可以抑制人骨肉瘤细胞株MG63和U2OS的增殖和迁移,并诱导它们凋亡和向成骨细胞分化。Objective To investigate the effects of BMP-2 and BMP-6 on osteosarcoma cell line MG63 and U2OS. Methods The two osteosarcoma cell lines were infected with AdBMP-2, AdBMP-6 (as experimental groups), and AdGFP (as control), respectively. Abilities of their proliferation, transmigration, apoptosis and differentiation were detected by trypan blue exclusion test, transwell-room, acridine orange-ethidium bromide fluorescent staining, in situ cell death detection and the test of alkaline phosphatase activity respectively. Results In the experimental groups, the cell survival rates were significant decreased (P 〈 0. 01 ) ; the apoptosis indexes were significantly increased in a time-dependent manner (P 〈 0. 01 ) ; the cell numbers of transmembrane were decreased ( P 〈 0. 01 ) ; and the activity of alkaline phosphatase was increased gradually ( P 〈 0. 01 ). Conclusion The human BMP-2 and BMP-6 could inhibit the proliferation and migration, promote apoptosis, and reverse the malignancy of osteosarcoma cell line MG63 and U2OS.
