摘  要:目的:检测急性非淋巴细胞性白血病患者骨髓白血病细胞表面MHC-Ⅱ分子和B7共刺激分子的表达情况。方法:采集分离34例初诊或复发的急性非淋巴细胞性白血病患者骨髓细胞,其骨髓中原始细胞比例>70%。应用结合有荧光PE或FITC的单克隆抗体标记骨髓细胞,用流式细胞仪进行HLA-DR、CD80(B7-1)和CD86(B7-2)免疫标记检测。结果:34例患者除M3型外,HLA-DR抗原的表达均较高,其中M2型最高,为(80.14±9.07)%;CD80阳性率均很低,最高为M1型,为(6.34±1.12)%,其余均在1%~4%之间;CD86的表达高于CD80,最高为M5型,为(32.60±4.18)%,最低为M6型,为(12.34±2.36)%。结论:急性非淋巴细胞性白血病细胞表面共刺激分子表达以CD80减低或缺乏为主。Objective:To examine the expression of MHC-Ⅱ and costimulatory molecules on acute nonlymphocytic leukemia (ANLL) cells. Methods:Bone marrow cells contained more than 70% leukemia blasts were isolated from 34 patients with ANLL in primary stage or relapse stage. The samples after labeling with monoclonal antibodies conjuncting with PE or FITC analyzed HLA-DR, CD80( B7-1 )and CD86(B7-2) on these cells by FACScan. Results:Expression of HLA-DR was high in all but ANLL-M3 in 34 patients with ANLL, and ANLL-M2 was the highest, about (80. 14±9.07 )%. Costimulatory molecule CD80 was low about (6. 34±1.12)% on ANLL-M1 and 1%-4% in other groups. CD86 expressed (32. 60±4. 18)% in ANLL-M5 and ( 12. 34±2. 36)% in ANLL-M6. Conclusion : In costimulatory molecules of ANLL blast cell, CD80 ( B7-1 ) expression is mainly low or absent.
