摘  要:应用划痕标记染料示踪技术检测β-胡萝卜素对促癌剂TPA抑制NIH/3T3细胞间隙连接通讯(GJIC)功能的影响,同时对细胞膜流动性和细胞膜唾液酸含量的变化与TPA抑制细胞GJIC功能的关系,以及β-胡萝卜素对这些变化的作用进行了研究。结果显示:β-胡萝卜素对NIH/3T3细胞GJIC功能有促进作用,并对TPA抑制NIH/3T3细胞GJIC功能有一定的拮抗作用,随剂量升高上述作用表现为增强趋势;TPA抑制细胞GJIC功能的同时显著升高膜流动性和膜唾液酸含量,而β-胡萝卜素对TPA诱导的这些效应有拮抗作用。The effects of β carotene on the inhibition of gap junctional intercellular communication (GJIC) between NIH/3T3 cells by tumor promoter TPA and roles of membrane fluidity and membrane sialic acid were studied.The results showed that β carotene promoted GJIC between normal NIH/3T3 cells and protected the inhibition of GJIC by TPA in a dose dependent manner.In TPA group,the membrane fluidity and the membrane sialic acid increased siginificantly compared with control group.The above two indices in β carotene plus TPA group decreased siginificantly compared with TPA group.These results suggested that β carotene could protect the inhibition of GJIC by TPA,and the protective effects may be correlated with the changes of membrane fluidity and membrane sialic acid.
