实验性脑出血与活性循环因子异常

摘  要:目的探讨大鼠脑出血后活性循环因子内皮素、肿瘤坏死因子,血管紧张素Ⅱ,心房利钠多肽的生物学效应及对脑出血周围脑缺血损害的病理学意义。方法实验动物为Wistar大鼠,随机分为脑出血组24只,对照组23只,采用自体血注入法建立大鼠尾状核出血模型,分别于脑出血后4h、8h、12h、24h、72h,及7d取血,采用放射免疫分析法,进行外周血内皮素、肿瘤坏死因子,血管紧张素Ⅱ,心房利钠多肽含量测定。结果大鼠脑出血后4h血内皮素、肿瘤坏死因子,血管紧张素Ⅱ,心房利钠多肽均开始升高,8h达到高峰,12h后下降,7d后恢复至正常范围,8h的血清内皮素、肿瘤坏死因子、血管紧张素Ⅱ、心房利钠多肽测定值,与正常对照组比较有极其显著性差异(P<0.001)。结论活性循环因子内皮素、肿瘤坏死因子,血管紧张素Ⅱ,心房利钠多肽可能参与了脑出血后的缺血性病理损害。Objective To explore the pathology significances and biological effects to the bleeding vicinity brain of Endothelin, Tumor necrosis factor, Angiotensin Ⅱ and Atrial natriuretic polypeptide effected in laboratory cerebral hemorrhage. Methods Injecting self venous blood to produce the cerebral hemorrhage model in Wistar rat. Obtain the blood at 4h,8h, 12h, 24h, 72h, 7d after cerebral hemorrhage, and radio immunoassay was used to detect the serum endothelin, tumor necrosis factor, angiotensin Ⅱ and atrial natriuretic polypeptide. Results The level of serum endothelin, tumor necrosis factor, angiotensin Ⅱ and atrial natriuretic polypeptide was heighten at 4h after cerebral hemorrhage, get the peak level at 8h after cerebral hemorrhage, and then drop down at 12h after cerebral hemorrhage and recover to normal after 7d later. The levels of the four factors at 8h after cerebral hemorrhage are significantly higher than that in normal rat. Conclusions Endothelin, tumor necrosis factor, angiotensin Ⅱ and atrial natriuretic polypeptide may participate the pathology injury in rat cerebral hemorrhage.
