摘  要:目的研究瘦素在人脑动静脉畸形(brain afteriovenous malformation,BAVM)出血中的表达。方法应用半定量逆转录-聚合酶链反应(polymerase chain reaction,PCR)、免疫组化、Western印迹法等在转录和翻译水平检测瘦素和瘦素受体在BAVM中的表达。结果免疫组化法比较已出血和未出血BAVM的瘦素表达,发现瘦素和瘦素受体在已出血BAVM的畸形团中表达明显升高。半定量逆转录PCR和Western印迹法发现瘦素在已出血BAVM的畸形团上的转录和翻译表达明显升高。结论瘦素和瘦素受体在BAVM的畸形团中表达升高,而且在已出血的BAVM比未出血的畸形团高。这表明瘦素在BAVM的畸形团中存在特异性表达升高,瘦素可能参与BAVM的破裂出血和发展。Objective One of the common malformation (BAVM) is intracerebral hemorrhage. clinical manifestations of human brain arteriovenous The priority goal of modern therapy on BAVM is to lessen the morbidity and mortality of ICH. But the mechanism of BAVM rupture to hemorrhage is still unclear. Leptin, as a hormone of metabolism, was discovered based on researches of obesity, but recently its effects on other than obesity were focused. The goal is to investigate expression of leptin on human ruptured BAVM. Methods Samples of ruptured and non-ruptured BAVM specimens of nidus part and perinidal brain tissue were obtained during operations, then stored in -80℃ refrigerator. We analyzed their transcription and translation level of leptin and leptin receptor by different methods, such as RT-PCR, immunohistochemistry and western blot. Results Immunohistochemical slides showed higher expression of leptin and leptin receptor on nidus part of ruptured BAVM than that of non-ruptured. Compared with brain tissue around BAVM, nidus of BAVM had higher expression of leptin and leptin receptor both at transcription and translation level. Conclusions It suggests that leptin may play an important role in rupture and development of hemorrhage of BAVM. It opens a brand-new window on investigating the mechanism of BAVM hemorrhage and development.
