摘　　要：目的评价植入用缓释氟尿嘧啶的缓释效果和安全性,研究给药剂量和药物浓度的关系。方法42例乳腺癌患者接受植入用缓释氟尿嘧啶的术前、术中局部植入化疗,以不同的给药剂量及给药后不等的手术时间,观察其血液、组织浓度及用药后反应。结果(1)该药体内植入后具有良好的缓释作用,所有病例无明显不良反应。(2)植入局部和区域淋巴结可获得较高的组织药物浓度。结论应用植入用缓释氟尿嘧啶行乳腺癌局部植入化疗,安全性高,全身影响小,缓释效果佳;加大给药剂量,局部能获得更高的治疗剂量。Aim To assess the time-release effeet and safety of fluorouraeial sustained release for implant and to study the relation between dosages and eoneentrations. Methods 42 patients with breast eaneer were treated with fluorouraeial sustained release for implant totally preoperatively and at surgery at different time and dosage. The drug eoneentrations in blood and tissues and the patients response were monitored. Results First,the drug had good time-release effeet in vivo. All patients had no severe side effeet. Seeond, high drug eoneentrations were obtained in implanting region and local lymph nodes. Third, more higher eoneentration of dosage inerease. Conclusion Fluorouraeial sustained release for implant for local-implanted chemotherapy of breast cancer is safe and has little whole body reaction. Its time-release is satisfaetory, Higher dosage ean get higher local eoneentration.
