摘 要:背景:细胞凋亡及其调控基因在肿瘤(如胃癌)的发生、发展中的作用是当今研究的热点,有必要全面筛查胃癌相关凋亡基因。目的:研究胃癌发病过程中凋亡基因表达谱的变化。方法:利用人细胞凋亡相关基因cDNA芯片研究胃腺癌和癌旁组织中相关基因的改变。采用荧光定量聚合酶链反应(PCR)和组织芯片技术验证IGFBP4基因的表达情况。结果:在458个凋亡相关基因中,有17个基因在癌组织和癌旁组织中出现2倍以上的明显改变;4个基因表达增强,13个基因表达下调。荧光定量PCR和组织芯片验证结果与cDNA芯片相符。结论:发生改变的17个凋亡相关基因可能与胃腺癌的发病机制有关,并涉及整个凋亡途径,值得进一步深入研究。Background: Apoptosis and the genes regulating process have become a focus of interest in the study of cancer (such as gastric cancer) development and progression. It is necessary to screen apoptosis genes related to gastric cancer, Aims: To investigate the gene expression profiles of apoptosis related to gastric cancer. Methods: eDNA microarray relevant to human apoptosis was used to investigate and compare the gene expression profiles of gastric adenocarcinoma and the paracancerous tissue. IGFBP4 gene was selected to validate the testing of gene expression by comparing the results of eDNA microarray with the results of fluorescent quantitative polymerase chain reaction (PCR) and tissue microarray. Results: Of the 458 apoptosis-related genes, 17 genes were detected having≥2-fold difference of expression between the cancerous and paracancerous tissue, with 4 genes up-regulated and 13 genes down-regulated. The results of fluorescent quantitative PCR and tissue microarray were coincident with those of eDNA microarray. Conclusions: The 17 genes involved in the whole apoptosis pathway might be relevant to the pathogenesis of gastric adenocarcinoma, and further studies are warranted.
