摘  要:目的:探讨β-细辛醚对痴呆大鼠海马神经元蛋白质表达谱的影响。方法:SD大鼠随机分为正常时照组、模型组和β-细辛醚治疗组。模型组和β-细辛醚治疗组大鼠行脑立体定位注射术,模型组从大鼠右侧海马注入β淀粉样蛋白(Aβ_(1-40))5μg;治疗组:将Aβ_(1-40)和β-细辛醚(0.12%)混合后注入大鼠右侧海马。抽提各组大鼠海马神经元组织蛋白质,行双向电泳,采用PDQuest-7.1.1分析软件解析各组之间有明显差异表达的蛋白质。结果:β-细辛醚治疗组α-烯醇化酶(α-enolase)、钙/钙调蛋白依赖蛋白激酶(Ca^(2+)/Calmodulin-dependent protein kinase)的表达高于模型组,而尿激酶型纤溶酶原激活物(urokillasc plasminogen activator surface)、P53抑癌基因(P53 tumor suppressor)和未知蛋白(SSP6001)表达则低于模型组。Objective: To detect some proteins associated with the effect of β-asarone on promoting the survival of injured hippocampal neuronal cells in rats. Methods: The rats were divided into normal control group, Alzheimer disease (AD) model group and β-asarone treated group. The rats in the latter two groups received injection under stereotaxic apparatus and 5μg Aβ1-40, Aβ1-40 and β-asarone(0.12%) mixture was injected into the rats' right hippocampus respectively. The hippocampal neuronal tissues were sampled from rats in each group 30 days after injection, and the extracted proteins from these tissues were detected by using 2-dimensional electrophoresis. PDOuest-7.1.1 software was utilized to identify the differentially expressed proteins among these three groups. Results: There were five kinds of protein differendally expressed among the three groups. Compared with the model group, the expression level of α-enolase and Ca^2+/Calmodulin-dependent protein kinase were up-ragulated in the β-asarone treated group, while the expression level of urokinase plasminogen activator surface, P53 tumor suppressor and a unknown protein were down-regulated. Conclusion: β-asarone may promote the survival of injured hippocampal neurons in rats by virtue of up- or down-regulatlng the expression levels of the proteins mentioned above.
