摘 要:目的评价金雀异黄素(genistein,Gen)对兔视网膜中央静脉阻塞新生血管的产生是否具有抑制作用,并探讨其机制。方法采用氩激光直接光凝法封闭兔视网膜静脉建立视网膜中央静脉阻塞模型成功后1周,将兔分为2组,2组玻璃体内分别注射150μmol·L-1的Gen0.1mL和DMSO0.1mL,连续4d。用药1周后处死,行视网膜血管内皮生长因子(vascular endothelial growth factor,VEGF)免疫组织化学染色和视网膜HE染色,计数矢状面6μm视网膜切片中突破内界膜的血管内皮细胞核数。兔光凝前、光凝后当天、1d、7d、18d行眼底荧光血管造影。同时设立正常对照组。结果Gen治疗组视网膜发生新生血管的眼数较DMSO组少,但较正常对照组多。Gen治疗组、DMSO组和正常对照组平均每个切面突破内界膜的血管内皮细胞核数为12.18±4.11、18.19±5.51和0·34±0.02,视网膜VEGF免疫组织化学分数分别为1.02±0.15、1.94±0.31和0.33±0·12,3组之间差异有统计学意义(P<0.01)。结论Gen可抑制缺血性视网膜病变的血管新生,机制可能与Gen抑制视网膜VEGF的表达有关。Objective To investigate the inhibitory effect and mechanism of genistein on neovascularization of central retinal vein occlusion of rabbit. Methods Retinal vein occlusion of rabbit model was established with argon laser photocoagulation to retinal vein. After 1 week, rabbits were divided into two groups, and intravitreal injections of either 0.1 mL of 150 μmol . L-1 concen tration of genistein,or DMSO were given to one group,four days in succession. After 1 week,vascular endothelial growth factors(VEGF) were stained with immunohistochemistry method and retinas with HE method. The proliferated neovascular response was quantitated by counting the nuclei of endothelial cells of new vessels extending from the retina into the vitreous in 6μm sagittal cross sections. VEGF were determined on the cross sections after immunohistochem ical stain. Before laser and 0 day, 1 day, 7 days and 18 days after laser, fluorescence fundus angiography was performed to evaluate the retinal neovascularization. The normal control group was also eatablished. Results The number of neovascular nuclei per cross section of retina in Gen-treated group, DMSO group and control group was 12.18±4.11,18.19±5.51 and 0.34±0. 02,respectively;And the immunohistochemical score was 1.02± 0.15,1.94±0.31 and 0.33±0.12, respectively. There were significant differences in three groups (P〈0.01). Conclusion Genistein is an effective agent in the prevention and treatment of retinal neovascularization. This mechanism may involve in its inhibitory effect on VEGF.
