摘 要:研究了牛体外受精后早期胚胎体外发育时,向其基础培养液中加入牛磺酸对胚胎桑椹胚率、囊胚率和孵化囊胚率的影响,以探寻牛磺酸克服牛胚胎"体外发育阻滞"现象的作用。试验结果表明:以TCM-199+10%FBS为基础培养液(对照组),再加入7,14mM的牛磺酸(试验组),试验组与对照组的桑椹胚率分别为48.1%、47.4%和43.2%;囊胚率分别为26.4%、22.3%和21.0%;孵化囊胚率分别为21.8%、18.7%和0%。IVF后2细胞、4~8细胞及8~16细胞期,在基础培养液中分别添加7mM的牛磺酸时,桑椹胚率分别为48.7%、57.1%和52.0%,囊胚率分别为25.1%、30.7%和27.9%,孵化囊胚率分别为25.9%、28.6%和25.0%;而添加14mM牛磺酸组时,桑椹胚率分别为50.4%、56.4%和55.4%,囊胚率分别为26.5%、31.3%和27.7%,孵化囊胚率分别为27.5%、31.1%和29.9%。体外发育培养液中添加7mM牛磺酸可显著提高桑椹胚率和囊胚率(p<0.05),在4~8细胞期添加14mM牛磺酸最为合适。The effect of in-vitro-fertilized embryos cultured in basic medium with different taurine concentrations on the morulae rate, blastocyst rate, and hatched blastocyst were detected and compared in order to find the effect of taurine on avoiding in vitro development block of bovine embryos. The results showed that the morulae rates of the treatment groups with 7 and 14mM taurine were 48.1% and 47.4%, no difference with the control without taurine in TCM-199+10%FBS(43.2%) (p〉0.05). The blastocyst rates were 26.4%, 22.3% and 21.0% respectively, and the group with 7mM taurine was significantly higher than that of the other groups (p〈0.05).There was no emergence hatched blastocyst in the control groups. The hatched blastocyst rates of the treatment groups were 21.8%, 18.7% and 0%. The results from experiment I showed that when adding to 7mM taurine in the basic medium at the 2-cell period, 4-8 cell period and 8-16 cell periods, morulae rates were 48.7%,57.1% and 52.0% respectively. The blastoeyst rate were 25.1%, 30.7% and 27.9% respectively. The hatched blastocyst rates were 25.9%, 28.6% and 25.0% respectively. When adding 14mM taurine in the basic medium at the 2-cell period,4-8 cell period and 8-16 cell period, however, morulae rates were 50.4%, 56.4% and 55.4% respectively. The blastocyst rate were 26.5%, 31.3% and 27.7% respectively. The hatched blastocyst rates were 27.5%, 31.1% and 29.9% respectively. These showed that supplementation of 7mM taurine in the medium could improve the morulae rate and blastocyst rate significantly (p〈0.05), and adding to 14mM taurine at 4-8 cell period got the best effect.
