HSV-TK/GCV系统杀伤高、低转移卵巢癌细胞株旁观者效应的观察

摘　　要：目的探讨单纯疱疹病毒胸苷激酶/丙氧鸟苷(HSV-TK/GCV)系统杀伤高、低转移卵巢癌细胞时的旁观者效应及其与间隙连接蛋白(Cx)43表达的关系。方法比较经HSV-TK/GCV作用的高、低转移性卵巢癌细胞株Ho-8910pm和Ho-8910旁观者效应;检测两种细胞Cx43的表达。结果HSV-TK/GCV系统对Ho- 8910细胞产生明显的旁观者效应,而对Ho-8910pm细胞旁观者效应较弱;Ho-8910细胞的Cx43表达为7.12(平均荧光强度),而Ho-8910pm细胞的Cx43表达为1.34,两者相比,P<0.05。结论HSV-TK/GCV系统杀伤低转移卵巢癌细胞时旁观者效应强,与Cx43的表达水平有关。[ Objective ] To explore the relationship between connexin 43 （ Cx43 ） and bystander effect mediated by herpes simplex virus -thymidine kinase/ ganciclovir（ HSV -TK/GCV） approach on high and low metastatic ovarian cancer cell lines. [ Methods] The expression of Cx43 was analyzed in Ho - 8910pro and Ho - 8910 cells. Bystander effect was determined by the cells growth inhibitory rate. [ Results] There was much greater bystander effect in Ho -8910 than in Ho - 8910pm . The expression of Cx43 detected in Ho -8910 （MIF = 7.12） was more than that of detected in Ho -8910pm （ MIF = 1.34）. [ Conclusions] Bystander effect mediated by HSV - TK/GCV approach on Ho - 8910 cells is much greater ,which is related to Cx43 expression.
