摘  要:目的建立复方α酮酸片中5种氨基酸的含量测定方法。方法以2,4-二硝基氟苯(FDNB)为衍生化试剂,将片剂中的氨基酸定量衍生化,衍生化条件为在碱性溶液中60℃暗处反应1 h,制得5种DNP-氨基酸。色谱柱为Luna C18(250 mm×4.6mm,5μm);以甲醇-醋酸盐缓冲液系统梯度洗脱;检测波长为360 nm。结果衍生化产物性质稳定;5种氨基酸在各自的浓度范围内线性关系良好(r〉0.999 0);加样回收率为97.2%-102.8%,相对标准偏差均小于2.0%。结论该方法准确,简便,重现性好,专属性强,可用于该产品的质量控制。Aim To develop a method for determination of amino acids of compound α-keto acid tablets. Methods Derivated by 2 ,4- dinitrofluorobenzene,5 kinds of DNP-amino acids in the tablets were analyzed by RP-HPLC with an Luna C18 column(4.6 mm ×250 mm,5 μm) as stationaryphase,methanol/acetate buffer as mobile phase,The detection wavelength was at 360 nm and the flow rate was 1.0 ml ·min^-1. Results The average recovery of all amino acids were 97.2% - 102.8% and RSD of all amino acids were 2.0 %. Conclusion The method is simple, accurate, reliable and can be used for determination of amino acids in compound α-keto acid tablets.
