人胃癌 NKM-45 细胞株胞浆、胞核性激素受体含量测定及分析

摘　　要：作者采用葡聚糖包被活性炭法测定了人胃低分化腺癌ＮＫＭ－４５培养细胞中分离的胞浆及胞核ＫＣＩ抽提组分中的ＥＲ、ＰＲ和ＡＲ的含量。结果显示胃癌ＮＫＭ－４５细胞的胞浆及胞核抽提液中ＥＲ含量为２４．８ｆｍｏｌ／ｍｇ和２５．１ｆｍｏｌ／ｍｇ蛋白，而ＰＲ含量为１６．４ｆｍｏｌ／ｍｇ和１８．６ｆｍｏｌ／ｍｇ蛋白，ＡＲ均为阴性（＜１０ｆｍｏｌ／ｍｇ蛋白），说明ＮＫＭ－４５细胞为ＥＲ及ＰＲ阳性细胞，且ＥＲ和ＰＲ在该胃癌细胞的胞浆、胞核中均存在。作者认为胃癌可能是性激素依赖性肿瘤，适当的内分泌治疗可能对胃癌有效。The levels of estrogen(ER), progesterone(PR) and andogen receptor(AR) in the cytosol and nuclear KCI extractive fractions, which were separated from cultured NKM 45 cells (a human low differentiated gastric adenocarcinoma cell line), were determined with dextrancoated charcoal method(DCC method). The result showed that, in cytosol and nuclear extract of NKM 45 cells the levle of ER were 24.8 fmol/mg and 25.1 fmol/mg protein, the level of PR were 16.4 fmol/mg and 18.6 fmol/mg protein, but the AR were negative (<10 fmol/mg protein) indivedually. This result indicated that NKM 45 cell is ER and PR positive, the ER and PR existed in cyosol and nuclear extract of the gstric carcinoma cells. So the authors thought that gastric cancer is possibly sex hormone dependent and suitable endocrine therapy may be effective for gastric cancer.
