摘  要:目的探讨超声造影剂SonoVue联合超声辐照介导野生型P53基因对裸鼠胰腺癌的转染作用。方法构建野生型P53真核质粒表达载体pcDNA3.1+/P53,建立裸鼠胰腺癌皮下模型。将60只荷瘤裸鼠分成4组,第1组瘤体内注入造影剂+P53质粒,超声辐照瘤体;第2组注入P53质粒后超声辐照;第3组注射造影剂+P53质粒;第4组注入P53质粒。利用RT-PCR、western blot、免疫组化检测P53基因在组织中的表达情况。结果成功构建了P53真核质粒表达载体pcDNA3.1+/P53及建立了胰腺癌动物模型;超声辐照微泡造影剂可促进P53基因在胰腺癌中的转染及表达。第1组P53的表达量明显高于其他3组(P<0.05),第2组基因表达高于第3、4组(P<0.05),第3、4组间基因表达差异无统计学意义(P>0.05)。结论造影剂联合超声辐照可明显增加P53基因在胰腺癌中的转染,可作为促进P53基因转染胰腺癌的辅助方法。Objective To determine whether ultrasound mediated microbubble destruction could increase naked human wild type p53 plasmid delivery to pancreatic carcinoma in nude mice.Methods Sixty nude mice with pancreatic carcinoma induced by chemical intoxicants were divided into four groups.The p53 gene was cloned into a high expressing efficiency plasmid pcDNA3.1+ and p53 plasmid was injected into the pancreatic carcinoma with or without echo contrast agent,and it was exposed to ultrasound.RT-PCR,Western Blot and immunohistochemistry were to evaluate P53 expression level.Results Plasmid pcDNA3.1+/p53 and the pancreatic carcinoma model were successfully constructed.Expression of p53 mRNA was significantly high in nude mice transfected with p53 plasmid by means of ultrasound and contrast agent than others(P〈0.05).Compared with the nude mice without ultrasound irradiation,transgene expression in nude mice with plasmid DNA using ultrasound was enhanced significantly.Conclusions Microbubble contrast agent plus uitrasound exposure can significantly improve the transfection efficiency of p53,and it can be applied for pancreatic carcinoma treatment as a gene delivering method.
