摘 要:目的探讨酸性成纤维细胞因子(aFGF)对体外培养的大鼠肾小管上皮细胞生长的影响。方法通过酶消化法获取肾小管上皮细胞。将aFGF(低、中、高浓度)加入培养液中作为实验组,不加aFGF的作为对照组。分别在12、24、48和72h进行细胞计数及形态学观察。结果从大鼠肾成功地获取了肾小管上皮细胞。干预72h内,实验组和对照组的肾小管上皮细胞都有不同程度地增殖,12h内各组无明显区别,24、48、72h实验组与对照组相比差异有统计学意义(P<0.05)。结论aFGF有明显的促进肾小管上皮细胞存活与增殖的作用,存在剂量-效应和时间-效应关系。Aim To explore effects of acidic fibroblast growth factor(aFGF) on the growth of renal tubular epithelial ceils cultured in vitro. Methods Renal tubular epithelial ceils in rats were gained through digestion by pamcreatin, aFGF was added into cultures of renal tubular epithelial cells, as experimental groups. The number of cells was counted and the shape of cells was observed at 12,24,48 and 72 h. Results Renal tubular epithelial cells were successfully obtained from kidneys. After treated for 72 h, the renal tubular epithelial cells showed different proliferation in experimental and control groups. There was no obvious difference at 12 h, but there was statistical difference between the two groups at 24 h ,48h and 72 h( P 〈 0.05 ). Conclusion aFGF had very strong survival-promoting and proliferative effects on renal tubular epithelial cells. The effects correlated with the dosages and periods of aFGF.
