摘 要:目的观察血卟啉衍生物(hematoporphyrin derivative,HpD)光动力学效应(photodynamic therapy,PDT)对体外培养人鼻咽癌细胞株CNE2及其顺铂耐药株CNE2/DDP的生物作用;探讨PDT治疗多药耐药(mul-tidrug resistance,MDR)鼻咽癌的可行性。方法对体外培养的CNE2与CNE2/DDP细胞分别加入0.5、1.0、1.5、2.0、2.5、3.0、3.5、4.0、5.0μg/ml不同浓度的HpD,孵育4h,给予波长为630nm的半导体激光进行照射,照射的功率密度为20mW/cm2,照射能量密度分别为2、5、10、20J/cm2,继续孵育24h,用四甲基偶氮唑盐(MTT)比色法测定细胞存活率。结果血卟啉衍生物介导的光动力(HpD-PDT)对体外培养的人鼻咽癌CNE2和CNE2/DDP细胞均有杀伤作用,表现为HpD浓度、激光剂量的量-效依赖关系。激光能量为20J/cm2时,HpD对CNE2和CNE2/DDP细胞半数杀伤浓度分别为0.7μg/ml与1.5μg/ml,两者比较有统计学意义(P<0.01)。相同HpD浓度、激光剂量条件下,CNE2/DDP细胞存活率显著高于CNE2细胞(P<0.05)。结论HpD-PDT对CNE2和CNE2/DDP细胞均有杀伤作用,但CNE2/DDP细胞对其敏感性低于CNE2细胞,表明CNE2/DDP对HpD-PDT存在一定程度的交叉抗性,增加HpD浓度和增大激光剂量可以在一定程度上克服这种抵抗。Objective To investigate the potential use of hematoporphyrin derivative (HpD) for photodynamic therapy (PDT) in the nasopharyngeal carcinoma (NPC) cell line CNE2 and CNE2/DDP cultured in vitro. Methods CNE2 and CNE2/DDP cells were incubated in media contained various concentrations of HpD (0.5, 1.0, 1.5, 2. 0, 2. 5, 3. 0, 3. 5, 4.0 and 5.0 μg/ml) for 4 hours and illuminated with different light doses using a semiconductor laser at 630 um with an energy density of 2, 5, 10, 20 J/cm^2. We analyzed the survival rates of CNE2 and CNE2/DDP cells with MTT assay. After 24 hours incubation with Hpd-PDT. Results HpD-PDT could effectively kill the CNE2 and CNE2/DDP cells cultured in vitro. The killing degree was positively correlated with the concentration of HpD and the dosage of radiation. The IC50 of CNE2 and CNE2/DDP was 0. 7, 1.5 μg/ml respectively with significant difference (P〈0. 01), when they were exposed to 20 J/cm^2. The survival rate of CNE 2 / DDP cells was higher than that of CNE 2 under the same HpD concentration and radiation dosage ( P 〈0. 05). Conclusion HpD-PDT may effectively kill CNE2/DDP cells as well as CNE2 cells cultured in vitro,though CNE2/DDP cells are less sensitive to HpD-PDT than CNE2 cells. The cross-resistance of CNE2/DDP to HpD-PDT may be overcome by increasing HpD concentration and radiation dosage.
