摘  要:目的探讨细胞因子不同的组合方式对人骨髓AC133+富集细胞体外扩增的影响。方法常规富集AC133+细胞,按照细胞因子不同的组合方式分为2组,在不同细胞因子组合刺激下进行体外对照培养,观察AC133+富集细胞的扩增情况;应用甲基纤维素半固体培养法,观察不同组别培养7天、14天、21天AC133+富集细胞的集落形成情况及细胞凋亡率。结果观察组的细胞因子组合扩增倍数、集落形成数目于培养7天后明显高于对照组,P<0.01;细胞凋亡率于培养21天后低于对照组,P<0.05。结论观察组的细胞因子组合方式既维持了干细胞自我更新的能力又在很大程度上扩增了造血祖细胞,显示了充分的优越性。Objective To explore the influence of different cell factor combinations on the human bone marrow AC133 + cells ex vivo. Methods The MiniMACS was used to gather AC133 + cells. There are 2 groups: the trai and control group. Through the ex vivo comparison of cultivation under different cell factor combination stimulation, expansion of AC133 + selection cells is observed. Observe the clone formation of the AC133 + ells using Half - solid Methyl cellulose under different cell factor stimulation ( in 7 d, 14 d,21 d. ) and Observe the rate of cells apoptosis. Results The number the expansion folds and clones of AC133 + from trial group were significantly more than those from control in 7 d, 14 d ,21 d ( P 〈 0.01 ). The rate of cells apoptosis from trial group is lower than those from control in 21 d and 28 d, P 〈 0.05. Conclusion Through the 4-week short-term cultivation, Cell factor combination from the trail is a well choice combination for ex vivo expansion, the combination keeps the ability of hematopoietic stem cell s' ability:self duplication and supper suffer hematopoietic progenitor cells.
