摘  要:目的观察神经生长抑制因子(Nogo-A)及其受体(NgR)在脑梗死大鼠不同时间点表达的变化规律。方法将80只SD大鼠制成易脑卒中型肾血管性高血压大鼠模型后,随机分为脑梗死组和对照组(各40只),脑梗死组大鼠施行右侧大脑中动脉闭塞术,用免疫组织化学法和荧光定量RT-PCR法检测术后第1、2、4和8周脑梗死灶周围Nogo-A及NgR mRNA表达的变化规律。结果免疫组织化学显示脑梗死组Nogo-A和NgR含量在病灶周围先呈逐渐上升趋势,第4周达到高峰,较对照组显著升高(P<0.05),Nogo-A在第8周降低,与对照组无显著性差异(P>0.05);而NgR略微下降,但仍高于对照组(P<0.01)。荧光定量RT-PCR显示脑梗死组Nogo-A与NgRmRNA表达均先上升,在第4周达到高峰,与对照组差异有显著性意义(P<0.01),第8周Nogo-A mRNA表达下降仍高于对照组(P<0.01);NgR mRNA表达在第8周时与对照组无显著性差异(P>0.05)。结论脑梗死损伤可使梗死灶周围组织中Nogo-A及NgR表达增加,可能与脑梗死后神经再生障碍有关。Objective To investigate the changes of mRNA expression of neurite growth inhibitory factor Nogo-A and its receptor NgR during focal cerebral infarction. Methods The stroke-prone reno-vascular hypertension rat (RHR) model and the middle cerebral artery occlusion (MCAO) were established and the histochemical stain and quantitative fluorescent RT-PCR were employed to detect Nogo-A,NgR and their mRNA expression in the brain tissue surrounding the infarct. Results Nogo-A and NgR contents in histochemical staining were elevated gradually after MCAO,and reached the peaks at 4th week. At 8 th week,Nogo-A decreased and was notsignificantly different from that in the control group,while NgR slightly decreased and was still higher than that in control group( P 〈0.01). The mRNA expression of both Nogo-A and NgR in fluorescent quantitative RT-PCR was up-regulated in peri-infarcted area and reached peaks at 4th week. The expression of Nogo-A mRNA was down-regulated from 8th week,but still higher than that in the control group. The NgR mRNA expression was not significantly different from that in the control group at 8th week. Conclusions Both Nogo-A and NgR protein and mRNA expression were up-regulated following cerebral infarction. It may has something to do with disturbance of regeneration of nerves after focal cerebral infarction in RHR.
