摘  要:目的:研究生物样品中盐、酯、蛋白质等成分的变化对固相微萃取-气相色谱质谱法检测氯氮平浓度的影响。方法:用固相微萃取-气相色谱质谱法测定样品中氯氮平(CLP)、洛沙平(内标),通过改变样品基质中氯化钠、醋酸乙酯、血浆的含量,观察盐、酯、蛋白质对CLP检测的影响。结果:随着样品中盐含量升高,CLP、洛沙平峰面积增大,方法灵敏度提高;随着样品中酯类、蛋白质含量升高,CLP、洛沙平峰面积减小,方法灵敏度降低。各组CLP、洛沙平峰面积保持相同的变化趋势,CLP与洛沙平峰面积的比值相对固定。结论:样品中盐、酯、蛋白质等含量的变化对固相微萃取-气相色谱质谱法检测氯氮平的灵敏度有不同程度的影响,但在选取合适内标的情况下并不影响定量分析的准确性。OBJECTIVE To study the influence of salt, ester, proteins in biological specimens on determination of clozapine concentration by Solid-phase microextraction- Gas Chromatography Mass Spectrometry ,(SPME-GC-MS). METHODS Clozapine and loxapine (internal standard) in these samples were detected by SPME-GC-MS, and the influence of solt, ester, proteins were observed by content variety of NaCl , ethyl acetate and plasma. RESULTS With the increased of content of salt in sample, peak areas of clozapine and loxapine increased and method sensitivity raised. With the increased of content of ester or proteins in sample, peak areas of clozapine and loxapine decreased and method sensitivity lowered. The peak areas of clozapine and loxapine had the same change tendency and peak area ratios of them remained almost constant in each group. CONCLUSION The changes of contents of salt, ester, proteins in samples will influence sensitivity of detection of clozapine by SPME-GC-MS, but the accuracy of quantitative analysis will not be influenced if suitable internal standard used.
