摘 要:目的优化胰岛细胞分离纯化的方法,提高胰岛产量、纯度和功能。方法本实验经胰管注射胶原酶、胰腺静止消化分离成年大鼠胰岛,葡聚糖离心纯化,获得胰岛细胞,进行记数和功能鉴定。结果纯化后胰岛收获量为7021±861IEQ/胰腺(IEQ=胰岛当量,一个胰岛当量为一个150μm直径的胰岛),纯度达86%;胰岛形态结构完整,胰岛素释放试验显示胰岛细胞功能良好。结论细节优化可以显著提高胶原酶消化、葡聚糖不连续梯度离心分离胰岛的效率。Objective To improve output, purification and function of islet cells by optimizing isolation and purification methods. Method Rat islets were isolated with in situ ductal collagenase distention, stationary digestion and dextran gradient separation. The islet cells were counted, and the function was identified. Results After purification, (7 021 ± 861)IEQ/pancreas were obtained, with an average purity of 86% islets. The isolated islets demonstrated a well-preserved cell ultrastructure, and well-responded to glucose stimulation. Conclusion Detail optimization could improve the effectiveness of stationary digestion and dextran gradient sepa- ration of islet cells.
