摘  要:目的:研究天麻水煎剂对家兔主动脉环的舒张作用及机制。方法:采用家兔离体主动脉平滑肌标本,以去甲肾上腺素(NA)预收缩主动脉环后给予不同剂量的天麻观察其张力变化,并探讨去除血管内皮及不同信号通路抑制剂对血管张力变化的影响。结果:天麻对血管环静息张力无明显影响,但不同剂量的天麻可使10-6mol/L NA预收缩血管产生明显舒张(r=0.85,P<0.001)。去除血管内皮、10-4mol/L左旋硝基精氨酸(L-NNA,NO合酶抑制剂)或10-5mol/L甲烯蓝(MB,鸟苷酸环化酶抑制剂)可减弱天麻的舒张作用,但吲哚美辛(环氧酶抑制剂)和普萘洛尔(β肾上腺素能受体阻断剂)无影响。另外,8mg/ml天麻水煎剂对无内皮细胞血管环NA和KCl的量效收缩反应明显压低,且NA和KCl的PD2分别由温育前(6.90±0.93)和(1.53±0.55)降低为(5.61±0.70)和(1.10±0.25)。结论:天麻水煎剂对主动脉的舒张作用是内皮依赖性的,并与内皮释放的NO有关;也与平滑肌细胞膜上的受体依赖性Ca2+通道和电压依赖性Ca2+通道有关。Objective: To study the vasodilation effect of the water decoction of gastrodia elata B1 (GEB) on rabbit thoracic aortic rings and the possible mechanism. Methods: Rabbit aortic rings were isolated and precontracted with noradrenaline (NA), and their responses to different concentrations of GEB were investigated. Then, experiments were carried out to assess the effects of endothelium removal and different signaling pathway inhibitors on GEB-induced relaxation. Results: GEB did not change the resting tension of rabbit aortic rings obviously, but it could result in an obvious relaxation in NA-precontracted aortic ring in a dose-dependent way ( r = 0. 85, P 〈 0. 001 ). The relaxant effect of GEB was significantly reduced by the removal of endothelium and by the treatment with 10^- 4 tool/ L nitric oxide synthase inhibitor Nw-nitro-L-arginine (L-NNA), or 10^-5 mol/L guanylyl cyclase inhibitor hethylene blue (MB), but not by the treatment with prostaglandin synthase inhibitor or blockage of adrenergic β-receptor ( Porp ). In addition, GEB ( 8mg/ml ) decreased the dose response curves of aortic tings to NA or KCI in the absence of endothelial cells, and changed the PD2 values for NA from (6.90 ± 0.93) to (5.61 ± 0.70) and for KCl from ( 1.53 ± 0.55 ) to ( 1.10 ± 0.25 ) respectively. Conclusion : GEB can relax isolated rabbit aorta in an endothelium-dependent way, with nitric oxide involved. The relaxation is also related to the blockage of receptor-dependent and potential-dependent calcium channels in the cell membrane of smooth muscles.
