摘 要:目的观察不同浓度的佛波酯(Phorbol myfismte acetate,PMA)和离子霉素(ionomycin,Ion)作为淋巴细胞激活剂对大鼠淋巴细胞表面分子表达的影响。方法用不同浓度的PMA和Ion刺激大鼠淋巴细胞后,使用流式细胞仪检测其CD3/CD4/CD8表达的变化。结果单独使用高工作液浓度的Ion(15、20μg/ml)对大鼠淋巴细胞表面CD4有显著下调作用,而中低浓度(2、5、10μg/m1)则影响不明显。单独使用低、中、高三种浓度(25、50、75ng/ml)PMA刺激,发现CD3/CD4/CD8无明显改变,分别以(2、5、10μg/ml)二种浓度的Ion联合高浓度PMA刺激后发现CD3/CD4/CD8也无明显改变。结论在低于10μg/m1的Ion和低于75ng/ml的PMA浓度条件共同刺激下不会引起表面标记物,特别是CD4分子表达的改变,故有助于进行淋巴细胞刺激相关试验时选择合适的淋巴细胞刺激物浓度。Objective To investigate the effects of different concentrations of Phorbol myfismte acetate (PMA) and ionomycin (Ion) upon molecules expression on the surface of rat lymphocyte. Methods Different concentrations of PMA and Ion were used to stimulate rat lymphocytes. The surface expressions of CD3/CD4/CD8 were measured by flow cytometry then. Results The surface CD4 expression decreased significantly in high concentration groups of Ion (15 and 20μg/ml), and did not change significantly in middle or low concent-ration groups (2, 5, and 10μg/ml). There was no significant change of surface expression of CD3/CD4/CD8 in low, middle and high concentration groups of PMA (25, 50 and 75 ng/ml). And the surface expression of CD3/CD4/CD8 did not change significantly when different concentrations of Ion were used combined with high concentration of PMA. Conclusions The surface expression of lymphocyte molecules changed little under the stimulation of Ion (at a low concentration up to 10μg/ml) combined with PMA ( at a concentration up to 75 ng/ml ). And dais will help to choose a suitable concentration of stimulator in the study of lymphocytes.
