摘 要:目的:探讨基质金属蛋白酶9(Matrix metalloproteinase 9,MMP-9)的表达和巨噬细胞的浸润在口腔鳞癌的侵袭和转移中的意义及相互关系。方法:采用免疫组化SABC法分别检测MMP-9和CD68(标记巨噬细胞)在42例口腔鳞癌和10例口腔正常组织的表达情况。结果:MMP-9的表达和巨噬细胞计数在口腔鳞癌中均显著高于正常组(P<0.05);肿瘤组织中MMP-9的表达及巨噬细胞计数与口腔鳞癌的淋巴结转移、TNM分期有关(P<0.05);而且它们之间呈正相关关系(γ=0.443,P<0.01)。结论:MMP-9和巨噬细胞可能在口腔鳞癌的侵袭及转移中发挥重要作用,其机制可能与巨噬细胞作用于肿瘤细胞并协助上调肿瘤细胞中MMP-9的表达有关。Objective: To investigate the significance and relationship between matrix metalloproteinase 9 (MMP-9) and infiltration of macrophage in the process of invasion and metastasis in oral squamous cell carcinoma (OSCC). Methods: The expressions of MMP-9 and CD68 were detected in 42 cases of OSCC and 10 cases of normal tissues by immunohistochemical SABC method. CD68 was used to detect macrophages specifically. Results: The level of MMP-9 expression and the ratio of macrophages in OSCC samples both were significantly higher than that in normal tissues (P〈0.05). The level of MMP-9 expression and the counting of macrophages were correlated with lymph-node metastasis and TNM stage (P〈0.05) they showed positive correlation (7=0.443) with statistic significance (P〈 0.01). Conclusion: Both MMP-9 and maerophages may play an important role in the process of invasion and metastasis of OSCC and higher level of MMP-9 expression in OSCC tumor may be resulting from the macrophage infiltration.
