喉鳞状细胞癌中Skp2和p27蛋白的表达

摘  要:目的探讨细胞S相激酶相关蛋白(S-phasekinase associated protein 2,Skp2)、p27蛋白与喉鳞状细胞癌(简称喉癌)各临床因素及预后的相关性。方法采用免疫组化SP法检测79例喉癌患者肿瘤组织的Skp2、p27表达。结果喉癌中Skp2高表达率(53.16%)显著高于正常喉组织(0%,P<0.05);喉癌Skp2蛋白低表达组的5年生存率(72.18%)显著高于高表达组(44.17%,P<0.01)。p27蛋白在喉癌和癌旁喉组织中的高表达率分别为30.38%和90%,差异具有显著性(P<0.05);喉癌p27蛋白高表达组的5年生存率(72.98%)显著高于低表达组(51.13%,P<0.01)。将Skp2和p27结合分析,Skp2高表达并p27低表达组的5年生存率最低,与另一组相比具有显著性差异(P=0.001)。结论Skp2蛋白通过降解靶蛋白p27可能在喉癌发生、发展中发挥重要作用。 OBJECTIVE To investigate the correlation of S-phase kinase associated protein 2 (Skp2) and p27 expression with the clinical factors and prognosis of patients with laryngeal squamou cell carcinoma (LSCC). METHODS The expressio of Skp2 and p27 in 79 specimens of LSCC and 1 specimens of adjacent normal laryngeal mucos tissues were evaluated by SP immunohistochemist methods. RESULTS The overexpression rate Skp2 was significantly higher in LSCC(53.16 %) tha in adjacent normal laryngeal mucosa tissue (0 % (P〈 0.05). There was a significantly higher 5-yea survival rate in reduced Skp2 expression grou (72.18 %) compared with that in Skp2 overexpressio group(44.17 %)(P〈 0.01). The overexpressio rate of p27 was significantly lower in LSCC(30.38 % than in adjacent normal laryngeal mucosa tissue (90 % (P〈 0.05). There was a significantly higher 5-yea survival rate in p27 overexpression group (72.98 % compared with that in reduced p27 expressio group (51.13 %) (P〈 0.01). Skp2 expression wa negatively associated with p27 expression. Patie with both Skp2 overexpression and reduced p2 expression revealed the poorest 5-year surviv rate (42.29 %), and the 5-year survival rate other patients was 70.05 %. There was a significa difference(P =0.001). CONCLUSION Th expression of Skp2 protein may lead to decreas p27 level in LSCC, indicating that it may play a important role in the pathway of carcinogenesis an progression of LSCC.
