摘  要:研究了文蛤多肽对体外培养的人肝癌SMMC-7721细胞的抑制作用,结果表明,经5.0μg/mL文蛤多肽处理的体外培养的人肝癌细胞(SMMC-7721)生长缓慢,倍增时间延长,细胞生长抑制率达89.4%;处理后的癌细胞形态发生明显改变,处于G0/G1期的细胞明显增多,而S期和M期细胞减少,出现明显的凋亡峰,凋亡率为22.3%.实验结果表明,文蛤多肽能有效地抑制体外培养肝癌细胞的增殖活动,可通过改变肝癌细胞的形态及细胞周期而明显抑制细胞的增殖.A peptide (named as Mercenene) was isolated from Meretrix meretrix Linnaeu and used to treat the human hepatocar: cinoma SMMC-7721 cells to investigate its effects on their proliferation, microstructure, and cell-cycle. The cell proliferation and the inhibition rates were determined by cell counting methods and MTT methods, respectively. The microstructures of the cells were observed by scan electron microscope. The cell cycle was analyzed by flow cytometry. It was found that the SMMC-7721 cells, after it was treated by 5.0 μg/mL of Mercenene, grew slower than the control, their doubling time were postponed and the inhibitory rate on the cell growth was determined to be 89.4%. In the mean time,flow cytometry analysis showed that the cell proportions in G0/G1 phases were significantly increased and those in the S and M phase were decreased markedly. The apoptosis rate was up to 22.3%. These results indicat that Mercenene can effectively inhibit the proliferation of the human hepatocarcinoma cell line SMMC-7721 in vitro and may act as an apoptosis inducer in its antitumor effects via inducing morphological changes and cell cycle arrest.
