摘  要:选取包埋剂海藻酸钠浓度为2.0%、3.0%、3.5%、4.0%,固定剂氯化钙浓度为2.0%、3.0%、4.0%、5.0%以及酶液与海藻酸钠溶液包埋比为50%、20%、10%、5%,设计正交实验,将从细菌HB-5中提取到的莠去津降解酶进行固定化,测定不同配比制成的固定化酶对莠去津的降解特性。结果表明,莠去津降解酶的最佳固定化条件为海藻酸钠浓度为3.0%,酶液与海藻酸钠的包埋比为5%,氯化钙浓度为4.0%。最佳固定化时间为4h,适宜的保存温度为4℃。The immobilized methods of free enzyme were studied. The free enzyme extracted from the strain HB-5 was first immobilized according to the orthogonal design, a L9(43) orthogonal array , whose three factors were sodium alginate, calcium chloride and proportion of sodium alginate and crude enzyme. The design's three levels were sodium alginate content of 2.0%, 3.0%, 3.5%, 4.0%, respectively, calcium chloride content of 2.0%, 3.0%, 4.0%, 5.0%, respectively and proportion of sodium alginate and crude enzyme of 50%, 20%, 10%, 5%. Subsequently the degradation of immobilized enzyme to degrade atrazine was detected. The results indicated that the free enzyme can be immobilized best under conditions of sodium alginate content of 3.0%, calcium chloride content of 4.0% and proportion of sodium alginate and crude enzyme of 5% for 4 h. Otherwise, the optimum conserved temperature was 4 ℃.
