摘  要:目的分析食管鳞癌与正常食管上皮细胞蛋白质的表达差异,获取鉴别两者的分子标志物。方法激光捕获显微切割技术分离食管鳞癌和正常食管上皮细胞的纯细胞,双向电泳和质谱的方法检测两者表达的差异蛋白。结果48个差异蛋白点,通过质谱鉴定出20种有意义的蛋白,其中11种蛋白如galectin7、14-3-3proteinε等在食管鳞癌细胞表达明显增高,9种蛋白如MTCBP-1等表达下调。结论激光捕获显微切割可以有效地解决组织异质性的问题;食管鳞癌和正常食管上皮细胞的2-DE蛋白质图谱具有明显的差异表达,提示食管鳞癌的发生是多种蛋白质功能共同作用的结果。Objective To search for the biomarkers for differential diagnosis between human esophageal squamous cell carcinoma cells and normal esophageal epithelial cells. Methods Laser capture microdissection was used to separate human esophageal squamous cell carcinoma cells and normal esophageal epithelial cells. Proteome alterations were observed using 2 dimensional polyacrylamide gel electrophoresis (2-DE) technique and electrospray ionisation tandem mass spectrometry. Results A total of 48 protein spots were discovered and 20 were identified as significant,in which the expressions of 11 proteins such as galectin 7, 14-3-3 protein were upregulated and 9 proteins such as MTCBP-1 downregulated in esophageal squamous cell carcinoma cells. Conclusion Laser capture microdissection is a key technique in proteomic study, which can effectively resolve the problem of irrelevant tissues. A differential expression of 2-DE proteomics was verified between human esophageal squamous cell carcinoma cells and normal esophageal epithelial cells, suggesting that a variety of proteins may be involved in the pathogenesis of esophageal squamous cell carcinoma.
