摘 要:背景与目的:探讨五氯酚钠诱发中国仓鼠卵巢细胞(CHO)DNA损伤和染色体畸变的效应。材料与方法:五氯酚钠对中国仓鼠卵巢细胞(CHO)进行梯度染毒,应用单细胞凝胶电泳试验和体外染色体畸变试验观察其诱发细胞DNA损伤和染色体畸变。结果:80μg/ml和160μg/ml五氯酚钠诱发细胞DNA链断裂损伤的频率与对照组相比显著升高(P<0.01),且随着剂量增加,其损伤程度加重;无论是否加入代谢活化剂,受试物均引起染色体畸变率的明显增加(P<0.01)。结论:五氯酚钠能诱发CHO细胞DNA损伤和染色体畸变。BACKGROUND & AIM: To investigate the in vitro effects of DNA damage and chromosome aberration induced by sodium pentachlorophenol. MATERIALS AND METHODS: Different concentrations of sodium pentachlorophenol were added to the culture medium for ceils of hamster ovary (CHO) . DNA damage and chromosome aberration thus induced were evaluated by using single ceil gel electrophoresis(SCGE, comet assay) and chromosome aberration test. RESULTS: The frequencies of DNA damage caused by sodium pentachlorophenol in the treatment groups with dosages of 80 and 160 μg/ml were significantly higher than the control group, and showed a dose-response relationship. Sodium pentachlorophenol also induced a significantly increased chromosome aberration rate both with and without adding S9 mix. CONCLUSION: Sodium pentachlorophenol could induce DNA damage and chromosome aberration of CHO cells.
