急性白血病细胞肺耐药相关蛋白的表达及其临床意义

摘　　要：目的:研究急性白血病细胞肺耐药蛋白(lung resistance-related protein,LRP)的表达,观察其表达率与化疗缓解率的关系。方法:利用LRP单克隆抗体,采用流式细胞术(FCM)分别测定15位正常对照和79例急性白血病(a-cute leukemia,AL)患者LRP的表达率,并分析其表达的临床意义。结果:初治急性淋巴细胞白血病(acute lymphoblastic leukemia,ALL)组LRP的表达率为(29.29±6.72)%,高于初治急性髓细胞白血病(acute myelogenous leukemia,AML)组的(21.46±5.23)%,P=0.0182;复发/难治ALL组LRP的表达率为(41.18±9.06)%,也较复发/难治AML组的(30.44±11.51)%高,P=0.0154。复发/难治组LRP的表达率均高于相应的初治组,P值分别为0.0175和0.0138。急性白血病细胞LRP(+)组的临床缓解率为30.8%,明显低于LRP(-)组的77.5%,P=0.0081。结论:复发/难治组LRP的表达率高于初治组,LRP过度表达与急性白血病患者的临床耐药及临床疗效密切相关。OBJECTIVE： To investigate the LRP expression in acute leukemia and the relationship between the expression and clinical remission rate. METHODS： The levels of LRP were detected by monoclonal antibodies and flow cytometry in 15 normal controls and 79 patients with acute leukemia （AL）, and the clinical significance of the expression rates wes evaluated. RESULTS： The expression rate of LRP in the initial acute lymphoblastic leukemia （ALL） group （ 29.29 ± 6.72 ） % was higher than that of the acute myelogenous leukemia （AML） group （21.46±5.23）%,P=0.018 2. The expression rate of LRP in the relapse/refractory ALL group （41.18 ±9.06）% was significantly higher than that of the relapse/refractory AMLgroup （30.44±11.51）%,P=0.015 4. The expression rates of LRP in the relapse/refractory AL group were significantly higher than those of the corresponding initial group, P was 0. 017 5 and 0. 013 8. The clinical remission rate of LRP＋ patients （30.8%0） was lower than that of LRP- patients （77.5%）, P=0.008 1. CONCLUSIONS： The expression rat of LRP in relapse/refractory patients is higher than that of initial patients. Overexpression of LRP in AL is significantly related to the clinical prognosis.
