PRL-3在结直肠癌中的表达及意义

摘　　要：目的检测PRL-3在大肠癌、癌旁组织及转移癌组织中的表达,初步探讨其与大肠癌发生发展、侵袭转移的关系。方法用免疫组化及荧光定量PCR方法检测大肠癌、淋巴结转移癌及相应癌旁组织中PRL-3在蛋白及转录水平的表达情况。结果PRL-3蛋白在淋巴结转移癌中的表达高于原发灶及相应正常癌旁组织中的表达(P<0.05),在大肠癌组织中PRL-3的表达与癌组织的浸润程度、淋巴结转移密切相关(P<0.05),癌组织与相应的癌旁组织相比差异则无显著性(P>0.05),其表达与癌组织分化程度差异也无显著性(P>0.05),在mRNA水平的检测也得到了与上述结果一致的结论。结论PRL-3的表达与大肠癌侵袭转移关系较密切,其表达水平在一定程度上提示大肠癌的转移潜能,PRL-3蛋白可作为大肠癌诊断、治疗及预后判断过程中的一种较有价值的病理学指标。Objective To detect the correlation between the expression of PRL - 3 and the biological behaviors of human colorectal carcinoma. Methods Immunohistochemistry and Real Time RT PCR were used to detect the expression of PRL - 3 in colorectal carcinomas and normal mucosa. Results The levels of protein and mRNA expression of PRL - 3 in metastatic lymph nodes were significantly higher compared with the matched primary lesions and normal tissues （P 〈 0. 05）. No significant difference in expression was found between normal mucosa and colorcctal carcinomas. The levels of protein and mRNA expression of PRL - 3 was significantly correlated with lymph node metastasis and invasion depth （ P 〈 0.05）, but no correlation with differentiation was found （P 〉 0.05）. Conclusion PRL - 3 gene plays an important role in invasion and metastasis of the colorectal carcinomas, strongly suggesting that the level of PRL - 3 can be used to indicate the potential of metastasis.
