摘  要:目的:研究人白血病单核巨噬细胞THP-1对人胃低分化黏液腺癌MGC-803细胞NF-κB核转位的作用。方法:采用免疫荧光法检测细胞核转录因子NF-κB核转位的情况。结果:免疫荧光分析发现,处理组NF-κB均有不同程度的核转位,以非直接联合培养组(CO)核转位作用为最强(28个核转位细胞),条件培养基处理组(CM)次之(15个核转位细胞),对照组核转位细胞数为2,各组之间比较,P<0.05。结论:THP-1细胞条件培养基处理、与THP-1细胞非直接联合培养能促进MGC-803细胞NF-κB核转位。OBJECTIVE: To investigate the nuclear transloeation of NF-κB in human gastric cancer MC-C-803 cells affected by human leukemia monocyte/macrophage cell line THP-1. METHODS: The immunofluorescence detection was used to assess the nuclear translocation of NF-κB in MGC-803 cells. RESULTS: Immunofluorescence assay demonstrated that two treated groups had a different nuclear transloeation of NF-κB in contrast to the control. Moreover, the nuclear positive cells in CO group was 28, while 15 in the CM group and 2 in the control group. The differences between eeah group among the three groups were all significant, P 〈 0. 05. CONCLUSION : Conditioned medium of THP-1 and indirect co-cultured with THP-1 can promote nuclear transloeation of NF-κB in MGC-803 cells.
