摘 要:目的:利用生物信息学方法对survivin拮抗肽(SP-1~SP-8)进行理化性质的预测及相似性的比较,为拮抗肽的结构与功能研究提供预测和帮助。方法:应用ExPASy数据库分析短肽的Mr、等电点、疏水性等理化性质。利用Blastp和SCOP等数据库对survivin拮抗肽进行相似性比较。结果:经ExPASy数据库分析8条拮抗肽中有6条呈碱性,且大部分呈亲水性,半衰期由1.0~30.0h不等。Blastp分析表明有几种拮抗肽与RNA多聚酶,转录调控因子的活性中心部分序列相似。SCOP分析结果显示有几种拮抗肽与ras结合蛋白(RBD)和CAD结构域相似。Objective:To analyze and predict the physical and chemical characteristics of survivin antagonistic peptides ( SPs), and to compare its similarity with other known proteins and to provide some evidence for further research on antagonistic peptides' structures and functions. Methods:Integrated bioinformatics database ExPASy was used to analyze molecular weight (Mr), isoelectric point ( pI), hydrophobicity and half life of SPs. Blastp and SCOP database were used to analyze the similarity with other known proteins. Results:It was found that 6 of the 8 SPs were rich in basic amino acids,. most of which were hydrophilic and their half life was 1.0 - 30.0 hours. Similar sequences were found between several peptides and RNA polymerase, transcriptional regulator by Blastp, between several peptides and ras binding protein (RBD) and CAD domain by SCOP. Conclusion: Bioinformatical methods are helpful in the study of polypeptide and prediction of protein structure and function as well as drug designing.
